Nitric oxide (NO) can be produced from its physiological precursor L-arginine by the action of a family of enzymes known as nitric oxide synthases (NOS). The biological consequences of NO, however, are dependent on its mode of delivery. In this report, we have used quantitative ligand receptor binding to describe the interaction of NO with the soluble guanylate cyclase (sGC) in intact cells and enzyme extracts. Saturation analyses of NO-dependent binding of [3H]N(G)-monomethyl-L-arginine ([3H]NMMA) to intact monocytes and dendritic cells revealed a single high-affinity binding site with an apparent dissociation constant (KD) of 1-3 microM. The pharmacology of this binding site was consistent with that of sGC, which we recently described in monocytes. Maximum specific binding of [3H]NMMA to dendritic cells was significantly higher than that observed in monocytes, and was inhibited by 2 mM concentrations of L-NMMA, L-arginine, or the structural analogue L-canavanine. The absence of NO-dependent binding to sGC in BHK cell extracts indicated that only one species of NO-responsive sGC was present in these extracts.
